Many molecular techniques have been used in aiding the study of both the Natural and Engineered CRISPR-Cas9 systems.

Single Molecule Fluorescence Microscopy has given valuable insight into the binding location and mechanism of the Cas9 nuclease.

Bioinformatics laid the foundation for our understanding of the natural CRISPR system, specifically allowing researchers to infer the genetic and biological purposes of the CRISPR locus and thus the overall function of the cas9 nuclease.
